Hmm, good question. If it's not locked properly, then I think it will. And that suitcase's lock is already failing right? That's why the Niffler escaped in part 1. Newt should change the lock and probably put a spell that no animals escape it in case the lock is faulty.

It broke into the vault in the nomaj bank and into the jewelry store and the niffler babies broke out of their cage.

I'm sure that she did open it, but if Newt put the spell on it then nobody can open it..

I guess a spell works best if the other person or animal can't use magic, but if you're up against a witch or a magical beast the cards might not always be in your favor.
