Probably because muggles and wizards to have to live together and at Hogwarts, there are muggles who study there. They were probably told at an early age that their "kind" does not interact or tolerate muggles.

I'm not sure but maybe only a few. Hermione's a muggle but she has some family that are witches so that's probably the reason why she got a Hogwarts letter.

Sirius has always been a rebel and would not conform to rules etc. I think he's the exact opposite of his younger brother.

And I think these two may have had their differences growing up and that caused some friction between them but at least Regulus realized his mistake but it was too late.
