• Eddie Redmayne and Katherine Waterston have been cast as the leads.
• It's based on Rowling's book of the same name.
• It will be a trilogy of "megamovies".
• The screenplay is being written by J.K. Rowling, produced by Steve Kloves & David Heyman, and directed by David Yates.
• 1, 2 and 3 will hit cinema screens in 2016, 2018 and 2020, respectively.

Most of the main characters are already cast but casting is still on-going for the movie. (I wonder if they'll attend Comic-Con next year?)

Warner Bros. may have already cast the major roles in the “Harry Potter” spinoff, “Fantastic Beasts and Where to Find Them,” but the studio is still on the lookout for another young star. Warners is holding an open casting call for girls between the ages of 8 and 12 to play Modesty, a “haunted young girl with an inner strength and stillness,” the studio said. Modesty has “an ability to see deep into people and understand them.” Auditions are being held at 9 a.m. Saturday in London’s Excel Centre Hall. Applicants must be available for shooting dates between August and January. The “Fantastic Beasts” cast already has fans buzzing, with Oscar winner Eddie Redmayne starring as Newt Scamander, Ezra Miller (“The Perks of Being a Wallflower”) as Kredan, Katherine Waterston (“Boardwalk Empire”) as Porpentina, Dan Folger as Jacob and Alison Sudol as Queenie. David Yates is directing “Fantastic Beasts,” which is set to bow on Nov. 18, 2016.

I don't think you need to read the book to be able to watch this movie as technically the book is a "reference" for magical creatures. But it would still be a good idea to read it so more or less you have an idea of what to expect.
