Yes, because if I remember correctly, Valentine told him to kill himself or disappear and left. Luke left and then heard that Jocelyn had left too and was living in NYC so he changed his name and opened a bookstore to fly under the radar and be with her.

He was and once he knew Jocelyn had the cup, he would strike and try and get it from her. Valentine probably thought Luke knew because his connection with Jocelyn.
