They do seem to have an understanding. Cas gets Dean and knows that Dean can be stubborn and a little rough around the edges, but he knows that the Mark is causing him to act very impulsively even for Dean.

I really liked that moment..not because Cas was broken...but because it showed how much he cared about Dean. (We knew that already, but it's always nice to see it. )

Yes he did look heartbroken and what to see how Cas will help Sam in saving Dean from being a demon.

It definitely highlighted their bond because as much as Cas cares abut Sam I feel like his bond with Dean is different like deeper and more meaningful
