Oh it wasn't actually funny, this was when Elena was finding out about her birth mother. And she smiles after saying she was a cheerleader because Elena was too and I think she liked that

 I think that maybe it looks like Elena was laughing because Isobel was a cheerleader, but Elena tried to be, but turned out to not be so good at it. __________________
