He's already lost her a few times though which does make me question what will be different this time to make it final...

Its got to be a possibility... its already been described as heartbreaking and the fact is her and Rory have left the Doctor twice or three times before now, and yet keep coming back, so if they want to write them out for good, then that may be a possible outcome...

I suppose, I just hope that if it were to happen, it would make the doctor realize that he can't save everyone. That he really is doomed.

Depends what the circumstances of it is... I mean it could be in the style of Renette from Girl in the Fireplace where he goes back to see her, but its just too late and she's died of old-age after having a really happy life with Rory. It would be sad, and uplifting at the same time. And maybe the Doctor could say to Rory that he (the Doctor) could go back and fix things and that time can be written and for Rory to reply, 'Not those times. Not one second. Don't you dare.' And the Doctor quietly leaves, sad that he'll never see his friends again, but happy they lived a full and happy life, a life he'd never be able to live. This could work well.

That would be heart breaking but realistic, I suppose. But I want Moffat to give us more screen time!!!

I don't feel like we've had enough closure on the Amy11 relationship yet.

If he's going to kill her, give us some more screen time.

Butting in, Karen mentioned she wants Amy to die which is just

heartbreaking but I think we should see it in perspective. Her playing Amy is brilliant beyond words and she wants nothing more than a good send off and if she thinks this is the best way to say goodbye, I guess we all have to accept that.

It doesn't mean we want it though

The other way I can see it happening is, by coincidence, they arrive at the library and the Doctor's (obviously) very quiet and sad and Amy/Rory want to know why and after an adventure there, they corner him and he's forced to tell them that he watched their daughter die here and is now living in a computer where they will never be able to see her again. Amy and Rory are so disgusted and angry that the Doctor A) Kept it from them and B) Let it happen, they leave the TARDIS for good...

So either the Doctor/Amy friendship ends or Amy or Rory die... thats my prediction...
