He knew Kira enough to know that she would never say ILY until and unless she meant it. So when the FC (as "Kira") said it back, he knew she was lying. When the fake Kira then said that she said it to make Odo feel better, he knew that was another lie. Kira wouldn't ever say she loved him, or be with him, unless she loved him.
When they did get together, Odo knew it was because she did love him back. Laas's visit gave him some doubts as to Kira's amount of love, but he tells Laas that he's choosing Kira. The woman he loves more than anything, even his people. And he knows Kira loves him back just as much.
